What is the Pipette
A pipette, also called a pipette gun, is a measuring tool that transfers liquid from the original container to another container
within a certain range. It is widely used in biology, chemistry and other fields.
Principle
The design of commonly used clinical micropipettes is based on Hooke's law: that is, the length of the spring extension is
proportional to the elastic force within a certain limit, that is, the volume of liquid in the pipette is proportional to the elastic
force of the spring in the pipette.
There are two physical principles of micropipette dispensing: dispensing with an air cushion and dispensing with positive
displacement of the piston without an air cushion. These two micropipettes with different principles have different specific
application ranges .
Instructions
1. Choose the right pipette
Air displacement pipettes are often used when pipetting standard solutions (such as water, buffer solutions, diluted salt
solutions and acid-base solutions), and liquids with high volatility, high viscosity and density greater than 2.0 g/cm A positive
displacement pipette is used for sample loading in polymerase chain reaction (PCR) assays. For example, when pipetting
15ul of liquid, it is best to choose a pipette with a maximum range of 20ul, and a pipette with a range of 50ul and above is
not accurate enough.
2. Set the pipetting volume
Adjust the pipette volume control knob to set the pipetting volume. When adjusting the pipetting volume, the scale should
be rotated to the scale exceeding the set volume according to the size of the volume, and then back to the set volume to
ensure the best accuracy of pipetting.
3. Assemble the tip
When using a single-channel pipette, align the nose cone of the adjustable pipette with the mouth of the pipette, and press
down gently and vertically to tighten it. When using a multichannel pipette, align the first row of the pipette with the first
nozzle, insert it at an angle, and rock back and forth slightly to tighten.
4. Pipetting
Make sure the pipette, tip and liquid to be pipetted are at the same temperature; then rinse the pipette tip 1-2 times with the
liquid to be pipetted, especially for viscous liquids or liquids with a density different from water. When pipetting the liquid,
immerse the tip of the tip vertically at a depth of 2-3mm below the liquid surface (it is strictly forbidden to insert the tip into
the solution), loosen the operating lever slowly and evenly, wait for the tip to inhale the solution, and let it stand for 2-3 seconds.
And stick it obliquely on the wall of the container to drain away the excess liquid on the outer wall of the suction head.
5. Placement of the pipette
After using the pipette, press down on the push rod of the tip with your thumb, safely withdraw the tip and adjust its capacity
to the marked maximum value, and then hang the pipette on the special pipette rack; When not in use for a long time,
it should be placed in a special box.
